Description of the facility
FORTH is the largest research institution in Greece, with six Institutes, among which, the Institute of Molecular Biology and Biotechnology (IMBB) at Heraklion, Crete Greece. IMBB hosts the Molecular Entomology lab (Vontas Group), which is equipped with all necessary molecular biology facilities to perform both classical (e.g., PCR, RT-qPCR, ELISA, Sanger Sequencing, LAMP) and modern (e.g., multiplex TaqMan, ddPCR, NGS, automated platforms) molecular diagnostics for vector monitoring (vector species/subspecies, pathogen and insecticide resistance), with several years of experience in the field.
Course overview
The course is aimed at students, early career researchers, and/or applicants looking to strengthen their research and surveillance capacity in the area of vector monitoring. It includes a theoretical and practical module. Theoretical module: Principles of PCR-, sequencing- and isothermal- based methods and their pros/cons. Experimental design, troubleshooting, analysis of data and interpretation of the results. Facility design and operations (good laboratory practice). Real-world scenarios of molecular Dx applications (species/subspecies ID, pathogen and insecticide resistance detection). Practical module: Hands-on experience in: Design of assays with relevant software, Preservation of samples, Homogenization and nucleic acid (DNA, RNA) extraction, Application of PCR (qPCR, melting curve, TaqMan, 3rd-gen ddPCR), sequencing (Direct sequencing and overview of NGS), isothermal methods (LAMP, isothermal PCR), sample-to-answer automated methods, Raw data analysis, Result interpretation and presentation.
